Genome-wide effect of constitutive IFN-signaling

Using wild type mice and mice deficient for either IFNb1 or IFN-receptor, we analysed the effect of the constitutive IFN on genome-wide expression To look for the role of constitutive interferon, we compared levels of expression of Interferon-stimulated genes (ISGs) in alveolar macrophages or liver in three mouse lines that is, C57BL6, IFNb1-KO, and IFN-receptor KO mice. Specifically, we isolated RNA from rested alveolar macrophages or liver and made library using Illumina TruSeq kit for the library construction. One replica of each sample is provided.

本研究以野生型小鼠(wild type mice)、IFNβ1基因敲除(IFNb1-KO)小鼠及干扰素受体基因敲除(IFN-receptor KO)小鼠为实验对象,分析了组成型干扰素(constitutive IFN)对全基因组基因表达的调控效应。为明确组成型干扰素的生物学功能,我们比较了C57BL6、IFNb1-KO及IFN-receptor KO这三个小鼠品系的静息肺泡巨噬细胞(alveolar macrophages)或肝脏组织中干扰素刺激基因(Interferon-stimulated genes, ISGs)的表达水平。具体而言,我们从上述样本中分离总RNA,并采用Illumina TruSeq建库试剂盒(Illumina TruSeq kit)完成测序文库构建。本数据集为每个样本提供一个重复样本。
