The genome sequencing of Elysia chlorotica

1. Genomic DNA was extracted from the whole body of an adult animal. Six libraries with short-insert sizes (170 bp × 2, 500 bp × 2, and 800 bp × 2) were constructed according to the standard protocol provided by Illumina (San Diego, CA, USA). Paired-end sequencing was performed for the six libraries on the HiSeq 2000 platform according to the manufacturer’s instructions. 2. Genomic DNA was extracted from a pool of ~1000 larvae (whole organisms). Five mate-paired libraries with long-insert sizes (2.5 kb × 2, 5 kb × 2, 10 kb × 1) were constructed according to the standard protocol provided by Illumina (San Diego, CA, USA). Paired-end sequencing was performed for the five libraries on the HiSeq 2000 platform according to the manufacturer’s instructions. 3. Genomic DNA was extracted from whole organisms of multiple adult Elysia chlorotica. Sequencing was performed on the PacBio RS platform.
