To assess the transcriptional capacity of the Human cytomegalovirus (HCMV), total RNA was isolated from from HCMV infected MRC-5 (human lung fibroblast) cells and reverse transcribed using SuperScript IV using oligo(dT) primers for the poly(A)-selected fraction and random primers for the rRNA-depleted fraction of the RNAs. cDNAs were sequenced using the Pacific Biosciences RSII platform.
