Myocardin (MYOCD) and MEF2 are essential transcription factors for the heart development and cooperatively regulate the expression of cardiac specific genes through their physical interaction. Actin-related protein 5 (ARP5) is a nuclear localized actin-like protein and involved in the regulation of muscle differentiation. We demonstrate that ARP5 binds to the N-terminus of cardiac isoform of MYOCD protein, which contains MEF2 binding site, and inhibits the interaction between MYOCD and MEF2. To determine the genes regulated by MYOCD-MEF2 and ARP5, we performed RNA sequencing using murine p19 embryonic carcinoma P19CL6 cells exogenously expressing cardiac MYOCD, MEF2C, ARP5, and N-terminal fragment of ARP5.
