We analyzed the gene expression changes occuring at the first 24h of EpiLC differentiation in embryonic stem cells in which a regulatory region driving expression of Tcf15 has been deleted. These changes has been compared with wild-type embryonic stem cells. Overall design: E14Tg2a (Wild type) and delta 0.7 (cells with the homozygous deletion of Tcf15 regulatory region;chr2:152,177,049-152,177,722) cells were cultured in serum-free 2iL medium. For the induction of differentiation, cells were plated onto fibronectin coated plates with N2B27 medium containing Activin, bFGF and KOSR. Samples were collected from 0h (2iL), 12h and 24h of differentiation in duplicates.
