资源简介:
We report the changes in the transcription profiles of the 4T1 breast cancer cells (control, RNF2 knockout (KO), and RSF1 KO), isolated and enriched from the tumor implanted in the 4th mammary pads by Fluorescence activated cell sorting, and the changes in the infiltrated NK cells. The transcription profiles were measured by RNA sequencing. We found that upon RNF2 KO or RSF1 KO, 4T1 tumor cells express multiple immune related genes, including chemokines, MHCII, and CD74 etc, which are otherwise repressed in cancer cells. We also found that the infiltrated NK cells in RNF2 KO tumors and RSF1 KO tumors express significantly higher levels of granzymes, compared to the NK cells infiltrated into control tumors, suggesting the activated status of these infiltrated NK cells. RNAseq examining the whole transcription profiles in control 4T1, RNF2 KO 4T1, and RSF1 KO 4T1 cells isolated and enriched from tumors implanted in the 4th mammary fat pads. RNAseq examining the whole transcription profiles in infiltrated NK cells in these tumors. ATACseq examining the chromatin accessibility in control, RNF2 KO, RSF1 KO and RNF2I53A/I53A mutant knockin 4T1 tumor cells enriched from corresponding tumors implanted into the 4th mammary pads of BALB/c. CHIPseq of Rnf2 determines the binding of Rnf2 to its target genes in the cultured murine breast cancer cells 4T1
