CrAssphages are one of the most abundant phages in the human microbiome, and there are close to 600 genomes identified computationally, but only four have been successfully cultured. Here, we identify and characterize three novel crAssphage species isolated from wastewater and infecting the bacterial host Bacteroides cellulosilyticus WH2. The novel species span two families and three genera which we have named Kehishuvirus winsdale (Bc01), Kolpuevirus frurule (Bc03), and Rudgehvirus redwords (Bc11). These three new species have podovirus-like morphology, and their approximately 100 kb genome has a consistent arrangement with known crAssphages. Crystal structure models of all 103 proteins from Kehishuvirus winsdale (Bc01), Kolpuevirus frurule (Bc03), and Rudgehvirus redwords (Bc11) were generated using Colabfold version 1.4.
交叉噬菌体(crAssphages)是人类微生物组中丰度最高的噬菌体类群之一,目前通过计算预测手段已鉴定出近600个基因组,但仅有4株成功实现体外培养。本研究中,我们从废水样本中分离得到3株新型交叉噬菌体,其感染宿主为解纤维类杆菌(Bacteroides cellulosilyticus)WH2。这3个新种分属2个科、3个属,我们将其分别命名为温斯代尔凯希病毒(Kehishuvirus winsdale,菌株编号Bc01)、弗鲁鲁尔科尔普病毒(Kolpuevirus frurule,菌株编号Bc03)以及雷德沃兹鲁奇病毒(Rudgehvirus redwords,菌株编号Bc11)。该3株新病毒均呈现短尾噬菌体(podovirus)样形态,其约100 kb的基因组结构排布与已报道的交叉噬菌体保持高度一致。 本研究利用ColabFold v1.4版本,为上述3株病毒的全部103个蛋白构建了晶体结构模型。
