Oncogenes NOTCH1 and MYC directly regulate HSF1 and other critical proteins of the stress-response pathway in T-ALL. This GRO-Seq experiment demonstrates that release from NOTCH1 inhibition results in upregulation of HSF1 and other key HSPs. Overall design: Nascent RNA was extracted from 10 million CUTLL1 cells using Trizol extraction (Invitrogen) according to the manufacturer’s protocol following the global run-on (GRO) reaction and BrUTP labeling. Nascent BrUTP-containing RNA was purified using BrdU beads (Santa Cruz) which was then used for library preparation. Libraries were sequenced on the Illumina HiSeq 2000 using 50bp single-read method. The experiment was performed at stready state (DMSO) and various time points post release from NOTCH1 signaling inhibition (γ-secretase inhibitor)
