Glucose-dependent insulinotropic polypeptide (GIP) is an incretin hormone secreted by enteroendocrine K-cells in the proximal small intestine. This study aimed to explore the function of human K-cells at the molecular and cellular level. Overall design: Organoids were generated using tissue from human duodenum. Venus fluorescent marker was inserted into GIP locus using CRISPR-Cas9 technology. Venus-positive cells from differentiated GIP-Venus organoids, and corresponding Venus-negative cells, were purified by flow-cytometry, and analysed by bulk RNA sequencing.
