Ribodepleted RNA-seq libraries from planarian tissues

In this study, we customized and optimized an RNaseH-based ribodepletion strategy and applied it to total RNA extracted from whole planarian worms, the outer epithelium (a.k.a., epidermis), and isolated blastema tissues. Our objectives were to create comprehensive RNA-seq libraries that: 1) contain less than 1% of rRNA contamination, and 2) include transcripts that are not polyadenylated, such as those encoding histone proteins and many noncoding RNAs.
