Therapeutic immune cell engineering with an mRNA : AAV-Sleeping Beauty composite system

We evaluated the insertion site profile of the MAJESTIC (mRNA : AAV-Sleeping Beauty) CAR generation method. Overall design: Genomic DNA was extracted from CARs and untreated T cells, and then a two step Splinkerette PCR was performed to generate an insertion site library that was sequenced by a Miseq sequencer.
