Bulk RNA sequencing of heads and antennae from mosquitoes fed with dengue, heat-inactivated dengue or a standard blood meal

This project investigated transcriptional changes in the brain and antennae of the African Tiger Mosquito, Aedes aegypti, following dengue virus (DENV-1) exposure via an artificial blood meal containing either live virus, heat-inactivated virus, or no virus. We performed bulk RNA-sequencing from 8 pools of 10 mosquito heads (appendages removed) and 8 pools of 60 antennae (30 pairs) collected from dengue-infected and control mosquitoes 14 days post-blood feeding. These RNA samples were extracted using the PicoPure Kit (ThermoFisher #KIT0204) combined with Trizol as described in PMID 33049200. Libraries were prepared with the Illumina Truseq stranded mRNA kit and sequenced on an Illumina NextSeq600 instrument.We subsequently implemented a single-mosquito cohort study design, collecting individual mosquito heads at 7-, 11-, and 14-days post-blood meal (DPBM). Individual mosquito heads underwent total RNA extraction with the PicoPure+Trizol method mentioned above. Carcasses were extracted with the Macherey Nagel RNA extraction kit. All samples were subjected to rRNA depletion with ZymoTech RiboFree library prep kit. Final libraries were prepared with the Illumina Truseq stranded mRNA kit skipping polyA capturing and sequenced on an Illumina NextSeq600 instrument.All reads in this studies are in paired-end (2x100bp) format and have been quality checked.For more information on this data please contact Umberto Palatini or Leslie B.
