GC-MS was utilized to analyze the metabolites in both the in vivo and in vitro experimental groups. In vivo, we analyzed VOCs in the blood of mice at 2, 4, 6, and 8 days after cell inoculation and on days 10 and 12 after tumor resection. We analyzed these variables based on the ion peaks in the resulting chromatogram and then evaluated their performance as predictors of CRC. In vitro experiments were performed by comparing changes in VOCs in HCoEpiC cells, the control group, the SW620 cell group, and the ATO+SW620 group.
本研究采用气相色谱-质谱联用仪(Gas Chromatography-Mass Spectrometry,GC-MS)对体内与体外实验组的代谢物进行分析。体内实验中,我们分别在细胞接种后第2、4、6、8天,以及肿瘤切除后第10、12天,对小鼠血液中的挥发性有机物(Volatile Organic Compounds,VOCs)进行检测。基于所得色谱图中的离子峰对上述变量进行解析,并评估其作为结直肠癌(Colorectal Cancer,CRC)预测标志物的效能。体外实验则通过对比人结肠上皮细胞(Human Colon Epithelial Cells,HCoEpiC)组、对照组、SW620细胞组及ATO+SW620组的挥发性有机物变化完成。
