This series is an updated dataset consisting of the Spec-seq and Methyl-Spec-seq samples for human CTCF with a bigger sequencing libraries and different epigenetic modifications. Each sample has replicate to gurantee the reproducibility for each measurement. Spec-seq is an intemediate throughput method to quantify the relative binding energy of some TF to thousands of DNA sequences with resolution down to 0.2kT. In this work, we utilized Spec-seq to study the recognition property ot CTCF, the genomic insulator, to its underlying core and upstream sequences, with different epigienetic modifications. Each sample has replicate to check for reproducibility. The state of epigenetic modification within CTCF's core binding site can be infered from the upstream barcoding positions. The EMSA separation work is performed in cold room, 12% Tris-glycine gel, 200V, 50mins.
