This project presents RNA sequencing (RNAseq) data from early postmitotic neurons isolated from the spinal cords of 7.5-day-old chicken embryos. The goal of this data collection is to conduct a comparative transcriptome analysis following the inhibition of semaphorin signaling during the development of dorsal horn neurons. This inhibition was achieved by overexpressing dominant negative forms of neuropilin 1 and neuropilin 2 receptors using an expression vector-based technology.In this process, the dominant negative plasmid was introduced into 4.5-day-old embryos through in ovo electroporation. After a survival period of three days, the transfected cells were isolated using fluorescence-activated cell sorting (FACS). Total RNA was then extracted from these cells for sequencing.There are three experimental groups in this study: 1) control GFP, 2) neuropilin 1 inhibited, and 3) neuropilin 2 inhibited. The results highlight the significance of semaphorin signaling in the early differentiation of dorsal horn neurons in the spinal cord.
