We report that the T cell specific adaptor protein (TSAd, encoded by the Sh2d2a gene) serves as a critical cell intrinsic molecule in CD4+Foxp3+ regulatory T cells to regulate cellular activation responses, mitochondrial translocation of Lymphocyte-specific protein tyrosine kinase (Lck) and immunoregulation following solid organ transplantation Overall design: CD4+CD25high regulatory T cells were FACS-sorted from Sh2d2a knockout and wild type mice and activated with 1 µg/ml anti-CD3 (clone: 145-2C11) for 2, 8 and 24 hours. Experiments were performed in duplicate conditions.
