NR study of the interaction between model membrane and lipid nanoparticles (LNPs) for gene delivery

LNP systems have played a critical role in the positive outcome of the COVID-19 vaccines. However, finding the optimised LNP formulations remains a significant challenge. It has been recognised that there is no universal solution. LNPs are internalized by cells through endocytosis and the subsequent endosomal escape process releases the payloads. Thus, high gene transfection and expression result from either a high LNP intake or an efficient gene release process in the cells, or both. These two processes are both related to the interaction between the LNPs with lipid membranes, either the cytomembrane or the inner membrane of the endosomes. Thus we propose the first study of how our LNPs and lipid membranes interact via a supported lipid bilayer model by neutron reflection. 3 days of Inter time is requested.
