The adult stem/progenitor cell maintenance in the prostate is regulated by a balance between BMP5 self-renewal signal and GATA3 dampening activity. Here we report that the transcription factor GATA3 controls basal stem/progenitor cell maintenance in the prostate by directly controling the expression of BMP5, a key effector of this activity. Wild type and PbCreGata3f/f prostate organoids were harvested at day 4 after 0, 2, 3 and 4 passages. Gene expression profiles were compared to identify candidate genes that lead to the amplification of the stem cell pool upon Gata3 inactivation in basal stem/progenitor cells from 8 week old adult mice.
