Genome occupancy of RNA Polymerase II and its phosphorylated forms were determined by ChEC-seq2 in S. cerevisiae. We test the dependency of RNAPII interactions on TFIIB, TFIIH Kinase, and the transcription factor GCN4. ChEC-seq2 with Rpb1-MNase and Rpb3-MNase (RNA Polymerase II). In addition, ChEC-seq2 was performed on components on the preinitiation complex, transcription activators, and coactivators. Changes in Rpb1-MN occupancy in response to TFIIB degradation, Kin28 inhibition (kin28is), and gcn4 deletion or mutation (gcn4-pd) were measured.
