This study describes the complete genomic sequence of an African swine fever virus (ASFV) mutant strain, GZ201801delta p54-150-164aa. The p54 protein of this isolate is characterized by a deletion of amino acid residues spanning positions 150 to 164. Ascertain ing the genomic sequence was aimed at elucidating the genetic basis of this mutation and its potential impact on viral replication and virulence. The ASFV was propagated on porcine alveolar macrophages (PAMs), followed by viral enrichment and extraction of the complete viral genomic sequence. Viral genomic fragments were amplified using the polymerase chain reaction (PCR) with specific primers and a high-fidelity reaction system. Subsequently, the sequences of these fragments were determined using the Sanger DNA sequencing method. These findings will contribute to a more comprehensive understanding of the genetic diversity of ASFV and the role of the p54 protein in viral biology.
