Mouse bone marrow-derived OSCAR+CTR+ osteoclasts were harvested from osteoclast-miRNA knockdown (OCmiR-KD) mice or osteoclast-miRNA intact (OCmiR-Intact) mice at 2w after anterior cruciate ligament transection (ACLT) by magnetic-activated cell sorting. The cells were lysated by Trizol reagent for total RNA extraction. The miRNA were profiled using the QuantiMir RT Kit (System Biosciences) Overall design: miRNA qPCR expression profiling. Equal amount total RNA from osteoclasts of three independent OCmiR-KD mice and three OCmiR-Intact mice were pooled for gene expression analysis.
