ScRNAseq of cardiac leukocytes from WT and Olfr2-deficient mice at day3 post AMI

We have employed a single-cell sequencing approach using PARSE Biosciences to investigate macrophage phenotypes post reperfused acute myocardial infarction (AMI) in Olfr2-deficient mice and wildtype controls. Macrophages are considered to be the main orchestrators in the inflammatory response after MI, contributing to adverse remodelling of the cardiac tissue. This is the first study to investigate the influence of the G-coupled Olfactory receptor 2 (Olfr2) on macrophage function and phenotype. For this study infarcted cardiac tissue was collected from Olfr2-deficient mice and wildtype controls 3 days following reperfused AMI. CD45+ cells were isolated by Fluorescence-activated cell sorting (FACS) and analyzed using scRNAseq. Sample-specific barcodes enabled computational demultiplexing after sequencing, allowing individual cells to be assigned to their respective animal. Separate gene expression matrices and metadata files were subsequently generated for each biological sample.
