Rewriter is an AAV-deliverable genome editing platform which utilizes a CRISPR guided reverse transcriptase to precisely edit genomic loci without double-stranded breaks. Plasmids encoding Rewriter protein and RNA components are transfected in HEK293T cells. Genomic DNA is extracted and high-throughput amplicon sequencing is used to screen the editing efficiency of various guide RNA constructs at different therapeutically relevant genes.
