Single cell RNA-seq analysis demonstrated dramatic changes in adult PgrCre/+Srfflox/flox endometrial cell populations compared to Srf flox/flox controls. Uterine Srf ablation caused myeloid immune cell infiltration, and differential expression analysis demonstrated that PgrCre/+Srfflox/flox stromal fibroblasts downregulated progesterone-regulated genes and growth response genes. Proliferating fibroblast populations shifted from G2-M phase-dominant in controls to S-G2 phase-dominant in PgrCre/+Srfflox/flox uteri. PgrCre/+Srfflox/flox epithelial cells overexpressed estrogenic and innate inflammatory genes. Overall design: Uterine tissues from two mice staged at gestation day 3.5 (3 days after finding vaginal plug) were pooled for each sample, and 2 samples per genotype (from 4 mice per genotype) were sequenced.
