Correlative cryo-SXT/ET workflow

During proposal 2023087704 we acquired extra datasets with controlled dose to evaluate if after cryo-SXT it would be possible to perform imaging with a TEM at cryo on the same cell region imaged with X-rays. The calculations we have done showed that about 20-30% of the dose is required for SXT. The cells imaged by SXT were then imaged by cryo-TEM at the Institut Pasteur confirming the dose calculated. Now we would like to acquire soft X-ray tomograms with the 2 lenses (25 nm and 40 nm), which have different efficiencies, on specific organelles to perform cryo-ET on these same specific organelles. The goal is to advance on demonstrating the usefulness of a correlative cryo SXT/ET imaging workflow.
