OsPAO5 mainly functions to oxidize spermine and produce H2O2,which finally affects mesocotyl elongation in rice. Meanwhile ethylene and polyamines synthesis crosstalk has been reported in competing the common substrate. To further confirm the relationship between H2O2 and ethylene in contributing to the mesocotyl length, we conducted a transcriptome comparison between pao5-1 vs NIP, ACC-treatment vs Mock (NIP) and H2O2-treatment vs Mock (NIP). Generally, the sterile grains of pao5-1 and NIP were sowed on a 0.6% agar medium containing 10 μM ACC or 10 μM H2O2, and cultivated for 7 days under absolute darkness environment (at 30 ℃ and 75% humidity). The 7-day-old mesocotyl tissues of pao5, NIP (Mock), ACC-treatment of NIP and H2O2-treatment of NIP were gathered to perform a transcriptome comparison.
