We found that active DNA demethylation is required for MAMP-triggered immunity to bacteria pathogen. Triple mutant plants deficient in ROS1, DML2, and DML3 (RDD) exhibited compromised immune responses such as induction of immune genes . Overall design: RNA-seq analysis was performed with 10-day-old seedlings or 4-week-old plants of WT, rdd with or without 100 nM flg22 treatment for 1 h, 3 h, 24 h, and priming . Three independent repeats were performed for RNA-seq analysis. DNA methylation profiles of 10-day-old seedlings of WT, rdd with or without 100 nM flg22 treatment for 1 h, 6 h, 12 h. Three independent repeats were performed for Whole-Genome Bisulfite analysis. Three independent repeats were performed for Whole-Genome Bisulfite analysis. For adult-plants, 4-week-old plants were hand-inoculated with H2O or 100 nM flg22 for 24 h. Two independent repeats were performed for Whole-Genome Bisulfite analysis.
