Study of Caenorhabditis elegans worms bearing the mutation daam-1(tm2133) revealed a variety of germline and growth phenotypes were closely linked to the daam-1 gene. However, attempts to knockdown daam-1 expression failed to recapitulate these phenotypes, and wild-type daam-1 failed to rescue, suggesting the causative mutations were in genes closely linked to daam-1. To identify such mutations, genomic DNA was purified from wild-type control strain N2 worms, and strain XA8002 worms bearing daam-1(tm2133) balanced by the transposition nT1[qIs51], and subject to whole genome sequencing.
