RNA-seq analysis to understand the regulation of genes by TET1 in mouse lungs and human bronchial epithelial cells. Overall design: An allergic airway inflammation model was established in Tet1+/+ mice and Tet1-/- mice using house dust mite allergen with appropriate controls. Lung RNA were extracted and compared using RNA-seq. In addition, endogenous TET1 expression was activated in human bronchial epithelial cells by dCasCRISPR for 24 hrs. RNA was extracted and compared between control and TET1 activated cells.
