To compare the inflammatory responses of WT and SIRPa KO macrophage, we performed a complete transcript profiling of WT and SIRPa-KO M1 macrophage using transcriptome sequencing as a discovery platform. SIRPa-KO mice and WT mice were kept under the same condition. BMDMs were produced from WT and SIRPa-KO mice followed by M1 polarization. RNA was then isolated from the same number of BMDMs. Overall design: In each group, 5 mice were used. The bone marrow-derived macrophages (BMDM) produced from WT and SIRPa-KO mice were treated with IFN?(20ng/ml) plus LPS (100ng/ml) in the presence of mCD47.ex for 6h. The M1 BMDM from 5 SIRPa-KO or 5 WT mice were mixed before RNA extraction.
