This study investigates the critical role of flagellin deficiency in antibiotic resistance mechanisms in Salmonella Typhimurium and Salmonella Choleraesuis, with direct implications for understanding the emergence of multi-drug resistant (MDR) S. 4,[5],12:i:-, a clinically significant monophasic variant. Our work demonstrates that the deletion of flagellin genes (fljB and fliC) in Salmonella enhances antibiotic resistance despite impairing biofilm formation. Intriguingly, residual biofilms in flagellin-deficient mutants exhibit significantly higher antibiotic tolerance, accompanied by upregulated efflux pump activity. These findings suggest that flagellin loss may drive MDR in S. 4,[5],12:i:- through dual mechanisms involving biofilm adaptation and efflux pump potentiation.
