Assay for transposase-accessible chromatin with sequencing (ATAC-seq) for shRenilla (control) or shClns1a transduced pTh17 cells restimulated with aCD3/CD28 for 24h. CD4+ T cells isolated from the spleen of WT mice were activated by plate-bound aCD3 (1 ug/ml) and aCD28 (1 ug/ml) in the presence of IL1b (20ng/ml), IL6 (20ng/ml) and IL23 (20ng/ml), aIL4 (5mg/ml). After 24 hours of activation Cells were transduced with shRenilla or shClns1a, then co cultured for another 24 hours to induce knockdown. Cells where then deattached, the media was changed, and samples were further cultured in the presence of IL1b (20ng/ml), IL6 (20ng/ml), IL23(20ng/ml), and aIL4 (5mg/ml) for an additional 96 hours.
