We carried out RNA-seq analysis to identify genes that are potentially regulated by PBX4 in hindlimb bud cells of the E11.5 mice by comparing gene expression in KO vs WT mice. Hindlimb buds were extracted from E11.5 embryos using forceps under a stereoscope. Total RNAs were isolated using TRIzol (15596-026; Invitrogen). High-throughput sequencing libraries were constructed using the NEB-Next Ultra RNA Library Prep Kit for Illumina.
