We performed single-cell RNA sequencing on CD45+ cells isolated from the lungs of C57BL/6 mice exposed to diesel exhaust particles via inhalation. The objective was to investigate which immune cells respond to particulate matter exposure. After DEP exposure, CD45+ immune cells were isolated from the lung tissues of C57BL/6J (wild-type) and Ccr2-/- (knockout) mice using fluorescence-activated cell sorting (FACS). The sorted cells were processed for single-cell RNA sequencing (scRNA-seq) to investigate transcriptional changes induced by DEP exposure. Control mice without DEP exposure were included for comparison. Libraries were prepared using the Chromium Next GEM Single Cell 3’ RNA kit v3.1 and sequenced on an Illumina NovaSeq 6000.
