To investigate the function of small heterodimer partner (SHP) in hepatocytes, we established Dox-inducible SHP overexpression iPSC cell lines and differentiated hepatocytes and cultured iPS-hepatocyte organoids (iHO). A comprehensive RNA-seq analysis was conducted. performed comparative gene expression analysis using data obtained from iHO vehicle and iHO dox. Overall design: After differentiation into iPS-derived hepatocytes, the cells were dissociated and seeded in matrigel semi-sphere. These cells were treated with 0,1 ug/mL doxycycline or vehicle. After about 2weeks of cultivation, grown organoids were harvested. Then total RNA was purified by RNeasy Mini. Then a library was prepared by Illumina TruSeq Stranded mRNA Sample Prep Kit. The data was obtained by Illumina NovaSeq. Comparison with two condition (Dox or Veh) were performed.
