Endothelial cells (ECs) are one of the major inductive cells in inflammation and keloid formation.The aim of this study was to identify gene expression patters of endothelial cells from keloid patients (KECs) comparing endothelial cells from normal patients (NECs). Immediately after the skin tissue excision, we performed CD31 positive ECs isolation. Keloid and normal skin tissue were gently minced with a GentleMACS Octo Dissociator with Heaters®. EC purification was performed using negative depletion with Anti-CD45 MicroBeads and Anti-Fibroblast for negative selection, and Anti-CD31-PE and Anti-PE Microbeads were used for positive selection for the CD31 positive EC by magnetic labeling with MicroBeads in MACS LS columns®. The purity of ECs as measured by flow cytometry was more than 85%. After the RNA extraction, Agilent Gene expression analysis was performed compared ECs isolated from keloid skin tissue (KECs) with ECs from normal skin tissue (NECs).Total RNA of KECs from 5 patients and NECs from 6 patients were analyzed.
