Deletion of Lats1/2 in adult kidney epithelia leads to renal cell carcinoma

We characterized the transcriptomes of both primary tumors and lung metastases tha developed in mice following deletion of Lats1 and Lats2 from Slc34a1-expressing cells as well as tumors arising from kidney allografts of and immortalized Lats1-/-;Lats2-/- cell line derived from the renal papilla. Overall design: 1. Primary tumors lung metastases were collected by dissection 4 months following injection of Slc34a1CreERT2;Lats1/2flox/flox mice mice with tamoxifen and processed for RNA-sequencing. 2. Proximal tubule cells were isolated by FACS from the kidneys of Slc34a1CreERT2;Lats1c/c;Lats2c/+;RosaTomato adult mice and processed for bulk RNA-sequencing. 3. Tumors were dissected from the kidneys of NOD/scid mice 6 weeks after injection of Lats1-/-;Lats2-/- cells under the renal capsule and processed for RNA-sequencing.
