Inflammatory response triggered by nerve injury plays important roles in the development of neurological disorders, such as neuropathic pain. The signaling events leading to inflammation in the nervous system remains to be better understood. Here, by deleting Dlk in sensory neurons, we found that DLK is required for neuronal intrinsic immune response to induce cytokines and chemokines including Il6, Il1a, Ccl2, Ccl7, and Ccl12 upon nerve injury. The neuronal immune response mediates immune cell infiltration in the dorsal root ganglia, and the microgliosis and astrogliosis in the spinal dorsal horn, thereby promoting inflammation in the spinal cord. To better understand the role of DLK in regulating neuronal intrinsic immune capacity at transcription level, we conducted SNI in WT and DLK cKO mice followed by RNA-Seq analysis.
