A combination of droplet based and plate based single cell sequencing was used to profile and compare all cell types in the mouse and human ovaries throughout the reproductive span. Overall design: For each sample, the ovary was dissociated and cells were size filtered to separate large oocytes from small oocytes and somatic cells. Large oocytes were sequenced using SmartSeq2 while the suspension of small cells was sequenced using 10X Genomics.
