This study aims to investigate the chemical constituents and activities of Menispermum dauricum. Separation and purification of its ethyl acetate fraction were performed using techniques such as preparative HPLC and silica gel chromatography, with compound structures identified by analyzing physicochemical properties and spectral data. The antioxidant capacity of the compounds was evaluated using the DPPH method, and their anti-breast cancer activity was assessed via the CCK-8 assay on MCF-7 cells. The structures of the compounds were identified based on their physicochemical properties and spectral data. The antioxidant activities of the compounds were evaluated by DPPH radical scavenging assay, and the antitumor activities were assessed by CCK-8 assay. A total of twenty-eight compounds were isolated from the ethyl acetate fraction of Menispermum dauricum and identified as p-hydroxybenzoic acid (1), N-methylcoraldine (2), fragransin B2 (3), methyl p-hydroxyphenylacetate (4), 2,6,2',6'-tetrahamethoxy-4,4'-bis(2,3-epoxy-1-hydroxypropyl) biphenyl (5), (-)-syringaresinol (6), (+)-dehydrofoliol (7), thalitine (8), mangiferin (9), syringic acid (10), 7-hydroxycoumarin (11), canthin-6-one (12), aquilegiolide (13), menisdaurilide (14), p-methoxyphenyl-propionic acid (15), vanillic acid (16), trans-N-p-coumaroyltyramine (17), cannabisin F (18), catechol (19), 4-methylpyrazole-3(5)-carboxylic acid (20), iseluxine (21), 3,4-dihydroxybenzoic acid (22), rel-5-(3S,8S-dihydroxy-1R,5S-dimethyl-7-oxa-6-oxobicyclo [3,2,1]-oct-8-yl)-3-methyl-2Z,4E-pentadienoic (23), N-trans-caffeinamide (24), acidcannabiamide D (25), melongenamide D (26), syringaresinol mono-β-D-glucoside (27).Compounds 5、7、9、11~14、23、24~27 were isolated from the genus Menispermum for the first time. Compounds 3, 8, 13, 24, and 26 exhibited potent DPPH radical scavenging activity, with IC50 values of 15.1 ± 1.4, 23.7 ± 2.1, 8.1 ± 1.7, 16.3 ± 1.1 and 23.5 ± 1.3 μmol/L. Compounds 2 and 12 showed inhibitory effects on MCF-7 cells, with IC50 values of 48.2 ± 1.4 and 40.3 ± 1.1 μmol/L.
