Ticks transmit viruses vertically and horizontally.The salivary glands, ovaries, and intestinal tract are vital in virus acquisition, transmission, and dissemination.From 2021 to 2023, ticks that parasitized animals (livestock such as sheep and cattle) were collected separately during the peak seasons of tick activity in Xinjiang, Inner Mongolia, Hubei, and Hainan, China.The salivary glands, ovaries, and midgut of the same tick group were dissected and pooled separately, creating one pool per tissue type for each tick group.Total RNA from each pool was extracted using the RNAiso Plus Reagent (Takara, Shiga, Japan) .RNA was fragmented and reverse transcribed into a strand-specific cDNA library using the VAHTS Universal V6 RNA-SEQ Library Prep Kit (Vazyme, Nanjing, China).Paired-end (2×150 bp) sequencing of each RNA library was performed using an Illumina NovaSeq 6000 System.
