To investigate the diversity and dynamics of clam (Meretrix petechialis) visceral mass microbiota during disease progression. We analyzed total bacterial communities using high-throughput sequencing of 16S rRNA gene during the vibrio immersion experiment. We have analyzed a total of 15 samples which represents 5 kinetic times (0, 1, 3, 4, 6 day post infection) in triplicates. For each sample, 16S rDNA amplicon libraries were generated using the 515F (5’-GTGCCAGCMGCCGCGGTAA-3’), 907R (5’-CCGTCAATTCMTTTRAGTTT-3’) primers targeting the variable V4V5 loops for bacterial communities. Paired-end sequencing with 250 bp read length was performed at the Novogene on the IlluminaHiSeq2500 platform. A total of 982,967 sequence reads were obtained from 15 libraries. After cleaning steps, 802,617 sequences corresponding to 1760 OTU were kept. Furthermore, singleton OTUs and sequences identified as chloroplasts or mitochondria were removed from the further analyses.
