Plasma proteome diversity: the overlooked albeit dominant contribution of immunoglobulins

By combining orthogonal separation methods, we charted more than 17,000 unique immunoglobulin (IgG1) antibodies in an individual human serum sample, exposing the wide-ranging contribution of immunoglobulins to plasma proteome diversity. Based on these data and considering also other (sub)types of immunoglobulins, we hypothesize that 5 – 8 million distinct clonotypic antibody proteins may co-occur in a single human serum sample. Here, we upload the Jupyter Notebook used to analyze the BiopharmaFinder output, which has been uploaded together with the RAW files to MassIVE. In addition, we provide Excel Files of manually curated dataframes for retention time alignment, as well as concentration determination. CSVs named D1_Fx_PTR are BPCs for reverse phase chromatograms (plotted in the jupyter notebook), files starting with MABMIX are chromatograms of intact mAb injections on the SCX column, while the 250820_SCX files are SCX-UV traces of the offline fractionated donor 1 IgG1 Fabs.
