sec6-4 cells expressing the immunoisolation bait Gap1GFPLTLM9 and the soluble secreted marker InvRFP from the inducible promoter Gals. Gap1GFPLTLM9 localizes to dot-like structures which is typical for the Golgi apparatus and endosomes in yeast. InvRFP is accumulating intracellularly at the restrictive temperature 37˚C. It is not secreted (shown also biochemically in Fig S3B).
