The goal of this project was to determine the differential expression of mRNA between two subtypes of Gbx2+ amacrine cells. These data will aid in distinguishing these two related, but distinct, amacrine cell subtypes by providing unique gene markers and physiological pathways. Two amacrine cell subtypes were labeled in mouse retina with fluorescent tags, tdtomato and GFP using Gbx2-CreERT2-IRES-EGFP; Ai9 transgenic mice. Mice retinae from P6-7 mice were dissected and Tdtomato labeled amacrine cells were purified by flow cytometry. Tdtomato-labeled amacrine cells were separated into two groups, GFP-negative (Cell type 1) and GFP-positive (Cell type 2), by flow cytometry. 4 biological replicates, originating from 3 different mouse litters, were collected. RNA was extracted from 750-2500 sorted cells. One GFP and one Tdtomato cell type sample will be collected from each animal. There was at least 1 male and 1 female sample used in this study.
