RNA-seq was performed in senescent BJ fibroblasts which had undergone stable JMJD6 or control knockdown since early young passages. Overall design: shRNAs targetting JMJD6 or control non-silencing sequences were stably transduced into young, non-senescent BJ fibroblasts. These lines were then cultured until reaching replicative senescence, at which point they were harvested for RNA-seq. Two separate JMJD6 targetting shRNAs were used, each into a separate cell line. RNA-seq tag density over gene bodies in JMJD6 knockdown cells were compared to the density in the control senescent sample.
