We investigated the role of NLRC5 in the endothelial cells (ECs) and in-vivo angiogenesis. The results showed that the deficiency of NLRC5 decreased angiogenesis in vitro and vivo.And the subcellular redistribution of NLRC5 was regulated by VEGFA-165. RNA-seq and Co-IP indicated the potential relationship of NLRC5 and STAT3. ChIP assay determined that the mRNA expression of angiopoietin-2 (Ang2) and cyclin D1 (CCND1) were orchestrated by the interaction of NLRC5 and STAT3. Overall design: RNA sequencing was performed after the following treatments: human umbilical vein endothelial cells were transfected with siRNA (siNLRC5 or negative control, 50 nM) for 48 h. After starvation for 12 h, VEGFA-165 (50 ng/mL) was used to stimulate HUVECs for 12 h.
