Purpose: Our objective is to profile and compare the cardiac endothelial cell transcriptome, identify and validate the candidate gene from the indicated experimental groups Overall design: Four different studies were conducted in C57Bl/6J mice: exercise trained vs. sedentary mice, high-fat diet (HFD) -induced obese vs. lean mice, aged (18 mo) vs. young (2 mo) mice and transverse aortic constriction (TAC) vs. sham -operated mice. Cardiac endothelial cells were isolated and sorted by FACS, and analyzed by RNA sequencing. cDNA library was synthesized using SMARTer Stranded Total RNA-Seq Kit V2 – Pico Input Mammalian (Takara Bio, USA) kit, The library quality was determined using bioanalyzer and sequenced using illumina NextSeq 550 System with the following specifications: 1 X 75bp, 50M single end reads.
