Stomata are key organs in plants and their development is influenced by a number of genes. The bgl23-D (bagel23-D) was a dominant mutant of Arabidopsis thaliana (EMS mutagenized Landsberg erecta) showing abnormal bagel-shaped stomata. The bgl23-D (Landsberg erecta background) was crossed with Columbia wild type and F2 lines where all F3 seedlings showed the mutant phenotype were selected. 50 of these homozygous F2 lines were combined and bulk population genemic DNA was subjected to whole-genome sequencing and analyzed by Mitsucal for mapping and identification of a mutation. The bgl23-D mutation was C to T substitution at the position of 3,220 bp in the AT1G02730 encoding ATCSLD5 (Arabidopsis thaliana cellulose synthase like D5).
