Downstream-of-gene (DoG) transcripts contribute to an imbalance in the cancer cell transcriptome

In this study, we reveal insights into the biogenesis of downstream-of-gene (DoG)-containing RNAs in human cancers. Examination of RNAPII and TOP1, catalytically engaged TOP1-sequencing, and nascent RNA sequencing in paired COADs and non-neoplastic colon tissue, SW480, HCT116, and FHC cells. RNAPII binding and nascent RNA sequencing upon TOP1 depletion in SW480 colon cancer cells.
